Diacylglycerols: the freshness clock hidden in the oil
Most of olive oil is whole, three-armed fat molecules. A small share are two-armed leftovers called diacylglycerols — and the way they slowly rearrange themselves over time makes them one of the most reliable, tamper-resistant freshness clocks a lab can read.
The bulk of olive oil is triacylglycerol — glycerol carrying three fatty-acid arms, the standard unit of any fat. But no oil is entirely tidy. A minor fraction arrives with only two arms attached: diacylglycerols, or DAGs. They form when the fruit builds fat or when a whole triglyceride loses an arm, and on their own they sound like a footnote. Their value is not that they exist, but that they slowly change shape in a way that quietly records the passage of time.
Two arms, two positions
The two fatty-acid arms of a DAG can sit in two different arrangements on the glycerol backbone: the so-called 1,2 form and the 1,3 form. Freshly pressed oil from healthy fruit is dominated by the 1,2 form — that is how the fruit lays it down. But the 1,2 arrangement is not the most thermodynamically comfortable one, so over time the arms migrate, and the oil steadily shifts toward the 1,3 form. Fresh oil is rich in 1,2; old, hot-stored or poorly made oil drifts toward 1,3. The ratio is, in effect, a slow clock.
Reading the clock
Because that migration is one-directional and steady, the share of DAGs still in the 1,2 form is a genuine freshness marker. In a good fresh extra virgin it runs high; some quality standards ask for at least about thirty-five per cent of the DAGs to be in the 1,2 form. Two things speed the drift toward 1,3: heat and high free acidity, both signs of tired fruit or careless handling. Refining pushes it hard too. So a low 1,2 share flags an oil that is old, badly stored, made from poor olives, or quietly cut with refined oil.
Why fraudsters hate it
Here is what makes DAGs so useful: unlike a printed date, the ratio cannot be back-dated. You cannot restore a 1,2 arrangement once it has migrated, and you cannot easily add fresh-looking DAGs without leaving other traces. The clock runs one way and keeps its own honest time. That is why serious buyers and some standards bodies lean on it — it exposes an oil that has been re-labelled with a flattering harvest date, or blended with a cheaper refined oil, no matter how convincing the front of the bottle looks.
